All target sequences can be amplified and visualized
If
too little amount of DNA exists, method of detection cannot be sensitive
enough. Because of that, a technique called PCR or polymerase chain reaction
is used. It consists of obtaining multiple copies from a DNA template.
This is accomplished by an enzyme named Taq-polymerase. This enzyme requires
two oligonucleotides complementary of the flanking regions of the sequence
of interest to yield acceptable specificity. This DNA molecules are designated
as primers.
Amplified
DNA of desired length can be visualized by using specific DNA dyes called
ethidium bromide or, alternatively, SYBR Green.
